The c u l t u r a l and n u t r i t i o n a l r e q u i r e m e n t s f o r t h e s y n t h e s i s of d-amylase by t h e mesophilic f u n g i , A s p e r q i l l u s n i q e r and A s p e r q i l l u s o r y z a e , i s o l a t e d from Nsukka soil were o p t i m i z e d . I n i t i a l pH and t e m p e r a t u r e optima f o r t h e p r o d u c t i o n of t h e enzymes i n t h e l i q u i d s t a t e w e r e 8.0 a n d 35O~fo r -A. o r y x a e a n d 5.0 a n d 45O~ for A. n i q e r r e s p e c t i v e l y . A n i q e r showed maximum p r o d u c t i o n of amylase i n a m i n e r a l s a l t medium contaihing W cocoyam, 1- 5% ( /v) Tween 20; 0 (V/v), and beanmeal, 1.0% (W/~).
